Horseradish Peroxydase (HRP) is a 40kDa protein that catalyzes the oxidation of hydrogen peroxide (H2O2) that, in the presence of substrate, results in a colored or fluorescent product. HRP is one of the most commonly used protein-markers in protein research and TMB (3, 3’, 4, 4’ – tetramethylbenzidine) is the most common substrate used in ELISA. Reaction between the acceptor and oxidized H2O2 will produce a blue colored solution. Once the desired intensity is obtained, the reaction is stopped with an acidic solution (STOP Solution). The reaction’s intensity can be monitored with a 96 well plate reader at 450nm.
